101 is a reference to college course structure.  Basically, you're learning about video games, so its like a course in the field.

What he said.  Usually '101' is the very first course you take in a college/university learning field.

Examples:

Computer & Info Science 101
Physics 101
Calculus 101

It's usually taken in your college Freshman year (your first year).
