I asked Dave Cox on Twitter regarding the co-op in this game. He replied to me with the following:

@TheUnifier " Zobek teams up with Gabriel in a few places but it's not a full co op. Zobek is AI controlled."

Just got a tweet from Cox:
  "Today I heard the waterfall music from Super Castlevania 4 fully orchestrated and it made me cry :)"

I asked him about the TGS trailer with Alucard, pretending not to know that the trailer wasn't from LoS  ;D Sneaky huh?
He didn't answer, unfortunately. I wanted him to at least say something like; "That's the japanese pitch that was scrapped" or maybe "I can't comment on that cause it's not made by Mercury Steam" (which would have led me to believe that the Alucardvania game was still in production.)
