I have not finished the game yet (I just defeated Brauner). But I already have plenty of favorite moments/moods that are as poignant to me as other memories from other favorites, Super CV4, SotN and CV:LoI.

Here are some of my LoS moments, list yours!
Dead Bog - the creepy mausoleum and the fight with the Swamp Troll at the graves

3 Towers - the art style, and running through while riding the Warg, with the water flowing everywhere.

Cornell's cutscene - love this character, his eye expressions, and voice.

Malphas Tower and staircase- loved the creepy gothic feel, and the red stairs. Oh, and the wraith that morphs from the torn curtain

Abbey Library - creepy abbey. awesome how the books can be destroyed and burned

Pause menu - Im always impressed at reading new monster backgrounds, and scrolls. Well written!

One of my favorite parts of the game is the very end of Chapter 5, Level 1 when you break down the last wall, run down the path, turn the corner and the music changes and you see the castle for the first time. Then you walk down the snowy path toward Wygol with the castle looming in the background...just an amazing scene.

The road to the castle, the stairway to the throne room, the astrologic puzzle, the Harry-potter-esqe chess was awesome ;D I think almost the whole wygol-castle part was awesome, the baba yaga segment was really cool and the last part of the necromancer abyss was also great. From the opening moments, the Mal`has tower is one of my favourites, I loved the color palette, graphis were breathtaking.

If I were to choose one single moment, it would be runing towards the castle.

Everything comes full circle

Nightfall in the Castle Hall was brilliant.

Absolutely. When we play this level in middle of the night in Paladin mode, we can feel the vampire's breath behind our neck.

My best part was the Level with Ogre just before Malphas tower. Best graphics, music and level design. The tower itself is quite a deception, though.

Boss fights are great, even Dracolich, just Satan is a little boring.

I just had that Skeleton crank moment in the sewers last nite too. Made me smile. That 1st fight with those skeletons was cool. Im impressed by their design, and that they don't all look the same.

I'm in the castle hall now, and love how you can pull the curtains. I didn't see it turn to nite yet, that sounds awesome! Well thought!

LOL i'm going through the game and looking at all the missed qte deaths and my favorite by far is the Carmilla one when you yank her onto the towers peak.
